HHV-6 subtyping is based on sequence variations within the U54 gene, which encodes the major capsid protein. There are four known subtypes of HHV-6A (variants A1 to A4) and three subtypes of HHV-6B (variants B1 to B3), which have been identified based on differences in nucleotide sequences. LaborantTM HHV6 Quantification Kit is used for quantification of the HHV6-A and HHV6-B with multiplex PCR amplification in a single tube using gene-specific primers. The kit allows to achieve RT-qPCR result in around 40 minutes and may vary according to the Ramp Rate of the device. The oligonucleotide set targeting human ß-globin gene functions as a control of the sampling, nucleic acid extraction and inhibition. All standards (HHV6-S E3, HHV6-S E4, HHV6-S E5, HHV6-S E6) have been meticulously designed and formulated to mimic HHV6-A/B positive human samples by making quantitative dilutions of HHV6-A/B gene fragments.
