42 dakikada Omicron (B.1.1.529) varyantının tespiti
Omicron VariAnt RT-qPCR Kit, COVID-19 hastalığına yol açan 2019-nCoV virüsünün, Omicron (B.1.1.529) varyantının tespitinde kullanılmaktadır. Kit içerisinde yer alan oligonükleotidler, Omicron varyantının varlığında ROX kanalında, yabanıl tip virüs varlığında Cy5 kanalında okuma alınmasını sağlar. Virüsün Spike (S) geni üzerindeki S371L ve S371F mutasyonlarını ROX kanalı üzerinden ayırt eder. Kit ile RT-qPCR sonucu, cihazın Ramp Rate değerine göre değişmekle birlikte, ortalama 42 dakikada elde edilebilmektedir. Çalışma materyali, nazofaringeal aspirat/lavaj, bronkoalveolar lavaj, nazofaringeal sürüntü, orofaringeal sürüntü ve balgam örneklerinden elde edilen nükleik asit izolatlardır. 2019-nCoV virüsünün ORF1ab, N, E ve S gen bölgelerini tek adımda ters transkripsiyon ve gerçek zamanlı PCR (RT-qPCR) ile tespit eder. İnsan RNase P geni ise, örnek alımı, nükleik asit izolasyonu ve reaksiyon engelleyici olası problemlerin tespiti amacıyla iç kontrol için çoğaltılır.
